Focus of Work

Academic Focus

My basic research interest is on the development of a genome-wide, CRISPR-Cas9-based screening platform to isolate regulators of the expression of immune checkpoint molecules on tumor cells and identify novel targets for anti-tumor immunotherapy. We demonstrated that acute myeloid leukemia cells up-regulated the expression of the immune checkpoint ligand PD-L1 upon interaction with T cells and the IFNR2-JAK1/2-STAT1-IRF1 axis was essential for this up-regulation. We showed that functional manipu...[Read full text]My basic research interest is on the development of a genome-wide, CRISPR-Cas9-based screening platform to isolate regulators of the expression of immune checkpoint molecules on tumor cells and identify novel targets for anti-tumor immunotherapy. We demonstrated that acute myeloid leukemia cells up-regulated the expression of the immune checkpoint ligand PD-L1 upon interaction with T cells and the IFNR2-JAK1/2-STAT1-IRF1 axis was essential for this up-regulation. We showed that functional manipulation of the oncogenes MYC and TP53 suppressed PD-L1 expression on leukemia cells and attenuated their ability to evade host anti-tumor immunity.[Shorten text]

Clinical Focus

My clinical research interest is on the application of an effective but less toxic method of stem cell transplant, called reduced-intensity conditioning transplant, to treat children with severe immune deficiency. We found that most patients tolerated this new approach better than the conventional approaches and had robust reconstitution of the immune system and correction of their underlying defects after the procedure.
