My basic research focus has been to investigate the role of the urokinase plasminogen activator system in tumor progression. In particular, my laboratory has focused on the role of urokinase (uPA) and its receptor (uPAR) in tumor growth, angiogenesis, and metastasis. In particular, my laboratory has focused on the role of urokinase (uPA) and its receptor (uPAR) in tumor growth, angiogenesis, and metastasis. As part of this work we have used animal tumor models extensively to evaluate the mechanism and tumor biology of this system in cancer progression. In parallel, I have spent over twenty five years working on discovering, developing and translating new oncology drugs into the clinic. During the course of this work, I have led multiple projects involving large groups of investigators focused on a variety of therapeutic targets and approaches including screening, candidate identification, pre-clinical development and early clinical trials and have worked on eleven INDs over the past fourteen years. This work has emphasized the need for improved animal tumor models, a case that I made when I came to Northwestern five years ago. We were fortunate to obtain philanthropic support that provided funding to generate the current PDX repository, which we are using in a number of drug development projects. I also have extensive experience in planning, implementing, and managing the evaluation of novel anti-tumor drug combinations and schedules.[Shorten text]
